چکیده

1 The cytokine transforming growth factor beta 1 (TGFB1) is implicated in male sexual 2 function. Previous behavioral studies show that Tgfb1 null mutant mice mount and 3 display limited intromission behavior with receptive females, but are unable to complete 4 successful copulation. The studies presented here explore the physiological basis for 5 sexual dysfunction in Tgfb1 null mutant males. Scanning electron microscopy revealed 6 that the surface of the penis in Tgfb1 null mutant males was abnormally coated in 7 superficial keratinized epithelial cells. There was a significant reduction in protrusion of 8 penile spines through the superficial tissue in Tgfb1 null mutant mice, and in some mice 9 the spines were almost completely embedded. Histological analysis revealed reduced skin 10 thickness in the penis of Tgfb1 null mutant males. Nerve fibres, endothelial cells, smooth 11 muscle actin, macrophages and neuronal and inducible nitric oxide synthase (NOS) were 12 present in similar abundance and location in Tgfb1 null mutant mice compared to 13 wildtype controls, however an increase in collagen I deposition was detected. Behavioral 14 studies revealed that Tgfb1 null mutant males undergo spontaneous non contact erections, 15 albeit at a reduced rate compared to control mice, and engage in less frequent genital 16 grooming activity. These studies suggest that Tgfb1 null mutation may adversely 17 influence copulatory behavior through effects on both altered structural integrity of the 18 penile skin, and impaired tissue compliance leading to erectile dysfunction. 19

Laminar Organization of Cerebral Cortex in Transforming Growth Factor Beta Mutant Mice

Transforming growth factor betas (TGF?s) are one of the most widespread and versatile cytokines. The three mammalian TGF? isoforms, ?1, ?2, and ?3, and their receptors regulate proliferation of neuronal precursors as well as survival and differentiation in neurons of developing and adult nervous system. Functions of TGF?s has a wide spectrum ranging from regulating cell proliferation and differ...

In Vitro and In Vivo Evidence that Thrombospondin-1 (TSP-1) Contributes to Stirring- and Shear-Dependent Activation of Platelet-Derived TGF-β1

Thrombospondin 1 (TSP-1), which is contained in platelet alpha-granules and released with activation, has been shown to activate latent TGF-beta1 in vitro, but its in vivo role is unclear as TSP-1-null (Thbs1(-/-)) mice have a much less severe phenotype than TGF-beta1-null (Tgfb1(-/-)) mice. We recently demonstrated that stirring and/or shear could activate latent TGF-beta1 released from platel...

Absence of integrin-mediated TGFβ1 activation in vivo recapitulates the phenotype of TGFβ1-null mice

The multifunctional cytokine transforming growth factor (TGF) beta1 is secreted in a latent complex with its processed propeptide (latency-associated peptide [LAP]). TGFbeta1 must be functionally released from this complex before it can engage TGFbeta receptors. One mechanism of latent TGFbeta1 activation involves interaction of the integrins alpha v beta6 and alpha v beta8 with an RGD sequence...

Effect of Vitamin C, as an Antioxidant, on Immobilization-Induced Changes in Sexual Behavior and Sperm Count in Male Mice

Sexual behavior in males is vulnerable to stress and it has been suggested that alterations in sexual behavior during stress is concomitant with spermatogenesis dysfunction. In this study, we investigated the effects of immobilization on sexual behavior and whether or not these effects are accompanied by changes in spermatogenesis process. The effect of antioxidant treatment on the sexual behav...

Changes in vascular reactivity of the coronary artery and thoracic aorta in the delta sarcoglycan null mutant mice

Introduction: Mutations in the delta sarcoglycan gene (d-SG) cause limb-girdle muscular dystrophy type 2F with structural and functional alterations in cardiac, smooth and skeletal muscle. The objective of the present study was to improve information about changes in vascular reactivity of the thoracic aorta and the coronary artery in the perfused heart of the d-SG-null mutant mouse model. ...
